5-Amino-1-methylquinolinium chloride is an enzyme overexpressed during skeletal muscle aging and is associated with repair of NAD+ pathway damage, dysregulated sirtuin1 activity, and increased muscle stem cell senescence. 5-Amino-1-methylquinolinium chloride promotes myoblast differentiation in vitro and enhances fusion and regeneration of muscle stem cells in aged mice. 5-Amino-1-methylquinolinium chloride is also a potent inhibitor of nicotinamide N-methyltransferase (NNMT), selectively binding to NNMT substrate binding site residues. 5-Amino-1-methylquinolinium chloride promotes myoblast differentiation in vitro and enhances fusion and regeneration of muscle stem cells in aged mice. Among them, the cytosolic enzyme nicotinamide N-methyltransferase (NNMT) has been newly discovered to regulate the levels of nicotinamide precursors required for NAD+ biosynthesis, and therefore plays a crucial role in regulating the NAD+ salvage pathway and cellular metabolism.
